Alterações na bioquímica hepática em cães com leptospirose aguda determinada por amostras do sorogrupo Icterohaemorrhagiae Ciência Rural

Foram testadas 120 amostras séricas de cães com títulos (<FONT FACE=Symbol>³</FONT>100) perante Leptospira spp. serovar Icterohaemorrhagiae para a determinação da atividade sérica de alanina aminotransferase (ALT), aspartato aminotransferase (AST), fosfatase alcalina (FA), gamaglutamiltransferase (GGT) e dos níveis séricos de colesterol total, triglicerídeos, proteína total, albumina, globulinas e bilirrubinas total, direta e indireta. Um grupo de 34 animais sem qualquer sintomatologia clínica e idade semelhante ao grupo testado foi utilizado como controle, tendo sido submetido às mesmas dosagens bioquímicas. An alternative for the preadsorption step in the paratuberculosis serodiagnosis: Mycobacterium fortuitum BJM

ELISAs for paratuberculosis employ a preadsorption step with Mycobacterium phlei to diminish unspecific reactions As M. fortuitum is one of the most frequent environmental mycobacteria, the purpose of this pilot study was to evaluate its use as an alternative for the preadsorption in ELISAs for paratuberculosis. Results suggest that M. fortuitum can be an alternative instead of or associated to M. phlei with comparable results (κ > 0.8) to conventional ELISAs using M. phlei as a preadsorption antigen.

Comparison of an in-house and a commercial enzyme-linked immunosorbent assay (ELISA) for diagnosis of paratuberculosis BJM

An in-house PPA-ELISA was compared to a commercial ELISA using a panel of 108 serum samples. In relation to commercial test, in-house assay presented 100% of sensitivity and 83.5% of specificity and a high concordance (kappa> 0.5), which demonstrates that the in-house assay is a valuable tool for use in developing countries.

Comparison of cefoxitin disk diffusion test and mecA gene PCR results for methicillin resistance detection in Staphylococcus intermedius group isolates from canine origin in Brazil BJM

The study evaluated cefoxitin disk diffusion tests breakpoints and their correlation to mecA gene PCR results for detecting Methicillin-resistant Staphylococcus intermedius Group (MRSP) isolates from dogs in Brazil. Agreement using proposed breakpoint (resistant < 30 mm) was encouraging. The current study reinforces that an epidemiological breakpoint can be established to predict presence of MRSP.

Comparison of serological methods for the diagnostic of Caprine arthritis-encephalitis (CAE) in Rio de Janeiro, Brazil BJM

Serum samples of 562 goats were tested for Caprine arthritis-encephalitis using agar gel immunodiffusion test (AGID) and enzyme linked immunosorbent assay (ELISA). Seventy-nine samples (14.1%) were reactive by both testes but 141 (25.1%) were reactive only by ELISA. ELISA, when compared to AGID, presented 100% sensitivity and 70.8% specificity.

Detection of Mycobacterium bovis DNA in nasal swabs from tuberculous cattle by a multiplex PCR BJM

Detection of tuberculosis in cattle relies on the intradermal tuberculin test (ITT), but a definitive diagnosis requires identification of the pathogen after the animal is slaughtered. DNA in nasal swabs from 50 cows was analyzed by m-PCR, targeting for the RvD1-Rv2031c and IS6110 sequences. M. bovis was identified in two of 34 tuberculous cows (5.9%). The use of mPCR of nasal swabs as an in vivo diagnostic tool for bovine tuberculosis is suggested.

First isolation of leptospires from dairy goats in Brazil BJM

Leptospires have never been recovered from goats in Brazil. Serum samples were obtained from 248 goats from Rio de Janeiro and from the seroreactive animals, urine samples were collected and processed for Leptospira isolation. A total of 52 positive reactions were observed, corresponding to 20.9% of the samples. The most prevalent reactions were to serovars Hardjo (36.5%), Shermani (30.8%), Icterohaemorrhagiae (9.6%), Grippotyphosa (9.6%), Autumnalis (5.8%), Castellonis (3.8%) and Bratislava (3.8%). Two strains of Leptospira sp. were isolated, both in the same region, but from different flocks. Presumptive identification based on serologic methods suggests those strains to be from Grippotyphosa serogroup.

Identification of Mycobacterium bovis Isolates by a multiplex PCR BJM

Isolates from suggestive bovine tuberculosis lesions were tested by a multiplex polymerase chain reaction (m-PCR) targeting for RvD1Rv2031c and IS6110 sequences, specific for M. bovis and Mycobacterium tuberculosis complex respectively. The m-PCR successfully identified as M. bovis 88.24% of the isolates.

Improvement of an in-house ELISA for bovine paratuberculosis serology in Brazil BJM

Paratuberculosis (Johne's disease) is a chronic enteritis in cattle caused by infection with Mycobacterium avium subsp. paratuberculosis (Map). In Brazil, few reports describe the isolation of the organism. The gold standard test for Map is its isolation from tissues or feces. Moreover, bacterial growth is slow and test results are available only after three to five months of incubation Furthermore, shedding of bacilli at levels detectable by fecal culture is irregular and does not occur during the early stages of infection, which compromises the sensitivity of this methodology. Interference of tuberculosis on the performance of ELISAs used in the diagnosis of paratuberculosis in cattle BJM

Forty-four cows from five herds infected with tuberculosis (TB) and without paratuberculosis (PTB), and 21 cows from a herd without either infection were studied. The cattle presented concordant results in both the skin test and gamma-interferon assay for TB and two commercial ELISAs for PTB. Animals were divided according to TB test results into Group A with 28 TB-infected animals, Group B with 16 TB-negative animals from infected herds, and Group C with 21 TB-negative cows from a tuberculosis-free herd (which were used as controls). ABSTRACT: Sheep breeding is an economic activity of great value for the Brazilian livestock industry, and infectious diseases can significantly compromise its productivity. The aim of this study was to detect Leptospira spp. in mixed breed sheep from a semiarid region of the State of Paraíba, Northeast Brazil. In total, 49 blood and renal tissue samples, as well as 25 urine samples, were collected from 49 sheep in the slaughter line. Molecular characterization of the first leptospires isolated from goats in Brazil BJM

Two Leptospira sp. isolates were obtained by the first time from goats in Brazil and characterized by sequencing rrs, rpoB and secY genes, PFGE and typing with monoclonal antibodies. Both isolates are identical and belong to Leptospira santarosai. Analysis of the rrs and the rpoB genes sequences revealed 100% identity between the goat isolates and the Bananal reference strain. When secY sequences of the two isolates were compared to each other, it was observed that they had identical sequences. However, when compared to that of the Bananal reference strain, there were 15 mismatches along the 549 bp secY sequence. Níveis séricos de uréia e creatinina em cães com leptospirose aguda determinada por amostras do sorogrupo Icterohaemorrhagiae Ciência Rural

Qualitative and quantitative analysis of bacteria from vaginitis associated with intravaginal implants in ewes following estrus synchronization Ciência Rural

ABSTRACT: This study evaluated the presence of vaginitis and the bacterial load associated with different intravaginal implants in ewes. Twenty-four Dorper and crossbred ewes were allocated into three groups and received intravaginal implant containing 0.3g progesterone (CIDR(r)), 60mg MAP or sponges without progesterone (CONTROL) for six days. Then, CIDR and MAP treated-ewes received 12.5mg dinoprost and 300IU eCG. Vaginal mucus samples were collected at four times: before device insertion, at the day of its removal, 24 and 48 hours after. The samples were cultured and the colonies were counted (CFU/mL) and identified. ABSTRACT: Reproductive efficiency is one of the key elements for any milk production system to be successful. In this regard, reproductive management must be planned properly and the identification of the main reproductive disorders that affect the herd is primordial for decision-making. The aim of the present study was to identify the main reproductive problems that affect dairy herds on Rio de Janeiro (RJ), Brazil. Data regarding breed, age, reproductive disorder occurrence, and breeding system were individually collected from dairy cows raised at 25 farms located at RJ. Seropositivity to Leptospira interrogans serovar Bratislava associated to reproductive problems without significant biochemical or hematological alterations in horses Ciência Rural

The objective was to study haematological and biochemical alterations associated to seropositivity to Leptospira interrogans serovar Bratislava infection in horses with reproductive alterations, such as neonatal deaths, embryonic deaths and abortions. A flock of mares with poor reproductive performance was studied. Eighty-two (58.6%) were seropositive (titre 200); 72 of those (87.8%) for Bratislava. Slight haematological and biochemical alterations were observed, being more frequent (P<0.05) in horses seropositive versus seronegative. In conclusion horses seropositive for Bratislava had no consistent, severe alteration in values for hematology and biochemistry serum. ABSTRACT: Leptospirosis is a zoonosis that is widely distributed in tropical countries. This infection is also associated with reproductive losses in livestock, which has a significant economic impact. The objective of this study is to investigate the seroprevalence and risk factors associated with leptospiral infection in commercial sheep flocks from the northwestern mesoregion of Rio Grande do Sul. Serum samples were analyzed in 319 sheep using the Microscopic Agglutination Test (MAT). At the time of sampling, an individual and herd questionnaire was carried out to investigate the risk factors.
